云南割手密是甘蔗育种中比较宝贵的野生种质资源。对68份云南割手密F2创新种质材料的9个主要工农艺性状进行主成分分析,并以前3个主成分为基础,分别作聚类分析和二维排序图。结果表明:68份云南割手密F2创新种质的变异系数为10.57%~35.30%,11月份理论含糖量和单产的变异系数超过30%,说明11月份理论含糖量和单产性状的变异潜力较高。主成分分析结果表明,单茎重产量因子、锤度糖分因子和丛有效茎含糖量这3个主因子提供了原始性状80.8292%的信息;在欧氏距离为10.00处将68份材料分为4类,各个类群具有不同的特点,在甘蔗杂交育种上可以根据不同类群的特点加以利用;二维排序表明云割F211-50、云割F211-56、云割F211-40、云割F211-66、云割F211-77、云割F211-10、云割F211-19、云割F211-85、云割F211-45、云割F211-37、云割F211-25、云割F211-23、云割F211-18等13份材料的3个主成分构成因子协调性最好,可在育种中加以利用。  相似文献

Genetic diversity among wild and cultivated barley as revealed by RFLP   总被引:4,自引:0,他引:4  
Genetic variability of cultivated and wild barley, Hordeum vulgare ssp. vulgare and spontaneum, respectively, was assessed by RFLP analysis. The material consisted of 13 European varietes, single-plant offspring lines of eight land races from Ethiopia and Nepal, and five accessions of ssp. spontaneum from Israel, Iran and Turkey. Seventeen out of twenty-one studied cDNA and gDNA probes distributed across all seven barley chromosomes revealed polymorphism when DNA was digested with one of four restriction enzymes. A tree based on genetic distances using frequencies of RFLP banding patterns was estimated and the barley lines clustered into five groups reflecting geographical origin. The geographical groups of land-race lines showed less intragroup variation than the geographical groups of spontaneum lines. The group of European varieties, representing large variation in agronomic traits, showed an intermediate level. The proportion of gene diversity residing among geographical groups (FST) varied from 0.19 to 0.94 (average 0.54) per RFLP pattern, indicating large diversification between geographical groups.  相似文献

[目的]蛀茎夜蛾Sesamia spp.是伊朗和其他国家(包括印度、巴基斯坦、斯里兰卡和日本)甘蔗上最具破坏性的害虫.蛀茎夜蛾幼虫钻蛀茎秆,对茎秆产生为害,降低重量和含糖量并降低甘蔗汁品质.应用抗性品种是一种有效的工具,对环境无不利影响.本研究旨在评价甘蔗商业品种对蛀茎夜蛾的抗性.[方法]在伊朗Ahwaz的Salman-Farsi Ago-industry Farms于2013-2014和2014-2015两个连续年份,采用随机区组设计进行5次重复试验.应用了CP69-1062,CP48-103,CP57-614,CP73-21,SP70-1143,IRC99-01,IRC99-02和L62-96 8个品种.在收获前,随机取20株完整的甘蔗茎秆进行蛀茎夜蛾的为害评估.记录受害茎秆百分比、被钻蛀的节间百分比(percent ofinternodes bored,IB)、出口孔的数目、活的蛀茎夜蛾的数目、每品种每公顷面积中蛾的繁殖量(mothproduction per hectare of each variety,MP).[结果]各测定参数在品种间均存在显著差异.从被钻蛀的节间百分比和蛾的繁殖指数判断,L62-96是最敏感的品种(2014年:14.58% IB,95 200 MP/ha;2015年:16.76% IB和111 300 MP/ha),其次是CP69-1062和CP48-103;CP57-614是这两年中抗性最强的品种(2014年:1.24% IB,8 400 MP/ha;2015年:1.02% IB,7 000 MP/ha).[结论]建议限制敏感品种的栽培,并应用其他控制措施,结合品种抗性,以治理敏感品种中的蛀茎夜蛾.  相似文献

Genetic diversity and structure of populations of the wild progenitor of barley, Hordeum spontaneum, from three countries, Israel, Turkey and Iran, in the Near East Fertile Crescent, are compared and contrasted. The analysis is based on electrophoretically discernible allozymic variation in proteins encoded by 27 shared loci in 2125 individuals representing 52 populations of wild barley. The results indicate that: (a) H. spontaneum in the Near East Fertile Crescent is very variable genetically; (b) genetic differentiation of populations includes some clinal but primarily regional and local patterns often displaying sharp geographic differentiation over short distances; (c) the average relative genetic differentiation (Gst) was 54% within populations, 39% among populations, and 8% between the three countries; (d) allele distribution is characterized by a high proportion of unique alleles (51%), and a high proportion of common alleles that are distributed either locally or sporadically; (e) discriminant analysis by allele frequencies successfully clustered wild barley of each of the three countries (96% correct classification); (f) a substantial portion of the patterns of allozyme variation in the wild gene pool is significantly correlated with the environment and is predictable ecologically, chiefly by a combination of humidity and temperature variables; (g) natural populations of wild barley are, on the average, more variable than two composite crosses and land races of cultivated barley. The spatial patterns and environmental correlates and predictors of genetic variation of H, spontaneum in the Fertile Crescent indicate that genetic variation in wild barley populations is not only rich but at least partly adaptive and predictable by ecology and allozyme markers. Consequently, conservation and utilization programmes should optimize sampling strategies by following the ecological-genetic factors and allozyme markers as effectively predictive guidelines.  相似文献

Elephantgrass (Pennisetum purpureum Schum.) and energycane (Saccharum spp. hybrid) are high‐yielding C4 grasses that are attractive biofuel feedstocks in the humid subtropics. Determining appropriate harvest management practices for optimal feedstock chemical composition is an important precursor to their successful use in production systems. In this research, we have investigated the effects of harvest timing and frequency on biomass nutrient, carbohydrate and lignin composition of UF1 and cv. Merkeron elephantgrasses and cv. L 79‐1002 energycane. Biomass properties under increased harvest frequency (twice per year) and delayed harvest (once per year after frost) were compared with a control (once per year prior to frost). There were no differences between elephantgrass entries in structural carbohydrates; however, elephantgrasses had greater structural hexose concentration than energycane for single‐harvest treatments (avg. 398 vs. 366 mg g?1), a trait that is preferred for biofuel production. Delayed harvest of energycane decreased structural hexose compared with the control (374 vs. 357 mg g?1) because nonstructural components accumulated in energycane stem as harvest was delayed. Frequent defoliation (2X) increased N, P, and ash concentrations (75% for N and P and 58% for ash) in harvested biomass compared with single‐harvest treatments. We conclude that multiple harvests per year increase the harvest period during which feedstock is available for processing, but they do not result in optimal feedstock composition. In contrast, extending the period of feedstock supply by delaying a single harvest to after first freeze did not negatively affect cell wall constituent properties, while it increased length of the harvest period by ~30 days in the southeast USA.  相似文献

The production and use of biofuels have increased rapidly in recent decades. Bioethanol derived from sugarcane has become a promising alternative to fossil fuel for use in automotive vehicles. The ‘savings’ calculated from the carbon footprint of this energy source still generates many questions related to nitrous oxide (N2O) emissions from sugarcane cultivation. We quantified N2O emissions from soil covered with different amounts of sugarcane straw and determined the direct N2O emission factors of nitrogen fertilizers (applied at the planting furrows and in the topdressing) and the by‐products of sugarcane processing (filter cake and vinasse) applied to sugarcane fields. The results showed that the presence of different amounts of sugarcane straw did not change N2O emissions relative to bare soil (control). N‐fertilizer increased N2O emissions from the soil, especially when urea was used, both at the planting furrow (plant cane) and during the regrowth process (ratoon cane) in relation to ammonium nitrate. The emission factor for N‐fertilizer was 0.46 ± 0.33%. The field application of filter cake and vinasse favored N2O emissions from the soil, the emission factor for vinasse was 0.65 ± 0.29%, while filter cake had a lower emission factor of 0.13 ± 0.04%. The experimentally obtained N2O emission factors associated with sugarcane cultivation, specific to the major sugarcane production region of the Brazil, were lower than those considered by the IPCC. Thus, the results of this study should contribute to bioethanol carbon footprint calculations.  相似文献

 Thirty-five populations of H. spontaneum from nine countries, encompassing almost all the known range of distribution of the species, Afghanistan, Crete (Greece), Cyprus, Iran, Iraq, Israel, Libya, Morocco and Turkey, were studied utilizing RFLP markers (21 probes with three restriction enzymes) distributed across all seven barley chromosomes in an attempt to unveil the genetic dissimilarities existing among them. UPGMA clustering, based on the Nei and Li (1979) similarity coefficient, produced a dendrogram where three clusters could be defined: two with a clear geographical distinction (Morocco and Cyprus) and another one grouping all the Asian/Middle Eastern populations, except for an accession from Iran that clustered separately. These results confirm our previous work and suggest that barley domestication could also have taken place outside the Fertile Crescent, particularly in Morocco. ACC(1-aminocyclopropane-1-carboxylic acid)合成酶是高等植物乙烯生物合成途径中的限速酶.根据已克隆的植物ACS(1-aminocyclopropane-1-carboxylic acid synthase)基因同源序列,设计简并引物,以甘蔗叶片总DNA为模板,通过PCR扩增,得到3条特异性强的扩增片段:Sc-ACS1为1 041 bp、Sc-ACS2为1 345 bp和Sc-ACS3为1 707 bp.将序列在GenBank核酸数据库进行同源性搜索,结果表明,3个片段均为ACS基因,推导编码的蛋白质序列分别包含326、242和310个氨基酸.其中,Sc-A CS1和Sc-ACS3同源性最高,核苷酸序列和蛋白质氨基酸序列分别有98%和96%同源,与禾本科植物玉米Zm ACS6、水稻OS-ACS2、毛竹等ACS基因家族也有很高的同源性,核苷酸序列同源性为88%-98%,蛋白质氨基酸序列同源性为73%-81%.甘蔗Sc-ACS2与水稻OS-ACS5在核苷酸和氨基酸序列上分别有91%和79%同源性,但与甘蔗Sc-ACS1和Sc-ACS3基因成员之间,氨基酸同源性分别只有45%和49%.系统进化分析表明,Sc-ACS1和Sc-ACS3基因与玉米Zm ACS6基因亲缘关系最近,而Sc-ACS2基因与水稻OS-ACS5基因亲缘关系最近.Southern杂交表明三基因在基因组中确实存在而且是多拷贝基因.三个片段已在GenBank数据库中注册,注册号分别为AY620985、AY620986和AY788919.  相似文献

【目的】蔗糖非酵解相关蛋白激酶2(sucrose non-fermenting 1-related protein kinase 2,SnRK2)在ABA信号转导途径和非生物胁迫响应中发挥重要作用。为进一步解析割手密SnRK2基因家族成员在干旱胁迫中的调控机制。【方法】该研究以全基因组数据为基础从割手密中鉴定SnRK2基因,并对其进行生物信息学分析和干旱胁迫下的表达分析。【结果】结果表明,在割手密基因组中共鉴定出11个SnRK2基因家族成员,命名为SsSnRK2.1-SsSnRK2.11,且这些基因不均匀的分布于8条染色体上。理化性质分析结果表明,SnRK2蛋白编码的氨基酸残基数为227~580,分子量为25 683.53~64 695.8 D,等电点为4.62~8.94,且均为亲水性蛋白。系统发育树可将其分为3个亚组,且同亚组中的保守基序基本相似,外显子数量以7~9个为主。顺式作用元件分析显示,SsSnRK2基因家族成员的启动子中含有多种激素类和逆境胁迫响应类的作用元件。不同组织转录组数据分析表明,割手密SsSnRK2s的表达具有组织特异性。qRT-PCR结果表明,所有的SsSnRK2基因均能不同程度地响应干旱胁迫。【结论】研究认为,SsSnRK2基因可能在割手密的干旱胁迫响应过程中发挥重要作用。  相似文献
